Dyeing Example

Description: 6-FITC can be used to label enzymes for detecting the enzymes immobilization distribution with green fluorescence.
Method: For preparation of 6-FITC-labeled enzymes.
1. FITC (1 mg/mL) solution is prepared using dimethyl sulfoxide is added dropwise into enzyme solution (5 mg/mL) prepared with 50 mM pH 9.0 carbonate buffer (prepared by adding 7.36 g NaCl, 1.06 g Na₂CO₃ and 7.56 g NaHCO₃ into 1 L deionized water).
2. Stir the mixed solution at 4℃ for 4 h.
3. Use 2 mL NH₄Cl aqueous solution (50 mM) to quench the reaction.
4. Finally, the solution is dialyzed in 50 mM, pH 7 phosphate buffer for 48 h at 4℃ for excess FITC removal.
5. Use a confocal laser scanning microscopy (Leica SP8 STED 3X) for image.
