PhosTAC7 is a heterobifunctional molecule named as a Phosphorylation Targeting Chimera (PhosTAC). PhosTAC7 can dephosphorylate the PDCD4 protein, FOXO3a protein, and tau protein by recruiting serine/threonine protein phosphatase 2A (PP2A). PhosTAC7 offers the advantage of selectively modulating the phosphorylation state of individual target proteins, making it a promising tool for research in cancer and tau protein-related neurodegenerative diseases (Alzheimer's disease).

Biologische Aktivität
PhosTAC7 (1-5 μM, 24 h) mediates ternary complex formation between Halo-PDCD4/tau and FKBP12(F36V)-PP2A A in HeLa cells[1][2].
PhosTAC7 (0.25-10 μM, 2-24 h) effectively induces the dephosphorylation of PDCD4/tau, and can simultaneously regulate multiple phosphorylation sites, with dose and time dependence in Hela cells[1][2].
PhosTAC7 (5-10 μM, 3-8 h) alters the phosphorylation kinetics of FOXO3a and resists the stimulation of growth factors depending on the activity of the PP2A phosphatase in Hela cells[1].
PhosTAC7 (0.5 μM, 24 h+ washout 24-48 h) still maintains a significant dephosphorylation effect even after the replacement of the fresh culture medium in Hela cells[2].
PhosTAC7 (1 μM, 1-3 d) indirectly accelerates tau degradation by promoting dephosphorylation in Hela cells[2].
