BODIPY FL-C16 is a BODIPY-labeled analog of Palmitic acid (HY-N0830), which serves as a fluorescent lipid tracer. BODIPY FL-C16 also acts as a ligand for liver fatty acid-binding protein (L-FABP) and intestinal fatty acid-binding protein (I-FABP) , with Kd values of 270 nM and 330 nM, respectively. BODIPY FL-C16 is rapidly taken up by cells, and after metabolic conversion to phospholipids, it is incorporated into the membrane structures of intracellular organelles and extracellular vesicles.

Biological Activity
BODIPY FL-C16 (1-7 μM; 15 min-5 h pulse, up to 24 h chase) metabolically labels a homogeneous subpopulation of ~80 nm small extracellular vesicles of endosomal/MVB origin in human Mel501, Me665/1, A375/M, HEK293T, and HeLa cells, with maximal secretion plateauing 6 h after a 2 h pulse, and the labelled vesicles showing high enrichment for CD63, moderate enrichment for CD81, and low enrichment for CD9[1].
BODIPY FL-C16 (0.5 μM; 5 μM L-FABP/I-FABP) has a low fluorescence quantum yield of 0.03 in PBS-CGM buffer, a high quantum yield of ~0.93 in methanol, and displays increased quantum yields of ~0.87 and ~0.72 when bound to recombinant rat L-FABP and I-FABP, respectively[2].
BODIPY FL-C16 (0.5 μM; ~1 min) binds to recombinant rat L-FABP with an apparent KD of 270 nM and to recombinant rat I-FABP with an apparent KD of 330 nM, occupying the fatty acid-binding site on I-FABP and one of two fatty acid-binding sites on L-FABP[2].
BODIPY FL-C16 (5 μM; 15 min at 37°C) extensively co-localizes with mitochondria and endoplasmic reticulum/Golgi, but not lysosomes, in fully differentiated human intestinal Caco-2 cells[2].
BODIPY FL-C16 (5 μM; 15 min at 37°C) is not esterified or incorporated into complex lipids in fully differentiated human intestinal Caco-2 cells within a 15-minute incubation period at 37°C[2].
