C14-306 is an ionizable lipid with a linear 306 amine core and a C14 alkyl tail. C14-306 enhances brain transfection efficiency. Lipid nanoparticles (LNP) formulated with C14-306 promote mRNA delivery, increase neuronal transfection rate, and facilitate Cre-mediated recombination in the brain, while reducing off-target expression in the liver.

Biological Activity
LNPs formulated with C14-306 (60 ng mRNA/20,000 cells; 24 h) efficiently and specifically transfect the neuronal cell line SH-SY5Y in vitro, exhibit extremely low transfection activity toward the brain microvascular endothelial cell line hCMEC/D3, and induce high uptake by neuronal cells[1].
