DOPE-PEG2000-MPG
DOPE-PEG2000-MPG (DOPE-PEG2000-Mal-Cys-MPG) is an amphiphilic lipid-PEG-peptide conjugate consisting of DOPE (1,2-dioleoyl-sn-glycero-3-phosphoethanolamine), PEG2000, and the MPG peptide, a cell-penetrating peptide derived from the HIV-1 gp41 fusion peptide and the nuclear localization sequence (NLS) of SV40 large T antigen. DOPE-PEG2000-MPG is used for intracellular delivery of siRNA, DNA, and other macromolecules.

Activité biologique
DOPE-PEG2000-MPG: surface modification of liposomes and nanoparticles for intracellular delivery of nucleic acids (siRNA, plasmid DNA, antisense oligonucleotides) and macromolecular drugs, particularly for gene therapy applications.
Structural composition:
(1) DOPE; a fusogenic phospholipid with two oleoyl chains that anchors into lipid bilayers and promotes membrane destabilization for endosomal escape;
(2) PEG2000; a hydrophilic polymer that provides steric stabilization and prolongs circulation half-life;
(3) MPG peptide; a bifunctional cell-penetrating peptide combining a hydrophobic domain (derived from HIV-1 gp41 fusion peptide for membrane interaction and crossing) with a hydrophilic domain (derived from SV40 NLS for nuclear targeting), enabling efficient cellular uptake of nucleic acid cargo through membrane destabilization and facilitating both cytoplasmic and nuclear delivery for gene silencing or gene expression.
