DSG-PEG2000-NHS is a conjugate composed of DSG, PEG chains, and terminal active esters (NHS). The NHS ester in DSG-PEG2000-NHS can rapidly covalently react with molecules containing primary amines (such as proteins, peptides, and small molecules modified with amino groups) to form stable amide bonds, enabling targeted modification of liposomes/nanoparticles.

The structure of DSG-PEG-NHS comprises three key components that work synergistically:
DSG: Acts as a hydrophobic anchor, responsible for firmly embedding the entire molecule into the membrane structure of lipid nanoparticles.
PEG chain: Acts as a hydrophilic spacer arm, forming a hydration layer on the particle surface, reducing non-specific adsorption and immune system recognition and clearance, thus prolonging the carrier's circulation time in vivo.
NHS: Acts as a reaction interface, reacting efficiently with molecules containing primary amino groups (such as antibodies and peptides) under mild conditions to form stable covalent bonds.
