Biological Activity
Guide (The following is our recommended protocol, which is provided as a guide only and should be modified to suit your specific needs).
Using CM-DiI to label lymphocytes to track their migration[1]
1. Preparation of stock solution
1.1 Dissolve 500 μg of CM-DiI in 250 μL of DMSO.
2. Preparation of working solution
2.1 Transfer 7.5 mL of Hank's Balanced Salt Solution pre-warmed to 37 °C into the DMSO solution containing CM-DiI, and mix for several seconds until the dye is evenly distributed.
3. Specific staining steps
3.1 Collect the overnight lymph fluid and determine the cell concentration. Take an appropriate number of lymphocytes, centrifuge at 4 °C for 10 min, discard the supernatant, and wash the pellet with HBSS.
3.2 Resuspend the washed cells in HBSS.
3.3 Immediately transfer the prepared dye working solution to the cell suspension and mix rapidly using a pipette.
3.4 Incubate the cells at 37 °C for 2 min, followed by an additional 2 min incubation on ice.
3.5 Centrifuge and wash the cells at 4 °C, resuspend them in 20 mL of Lactated Ringer's solution, place the cell suspension on ice, and perform injection within 15-30 min.
