CL-242817 is an orally available angiotensin converting enzyme (ACE) inhibitor. CL-242817 inhibits the conversion of angiotensin I to angiotensin II and has blood pressure lowering activity. CL-242817 effectively inhibits rabbit lung ACE in vitro (IC50=54.9 nM), angiotensin I (Al) induced contraction of isolated guinea pig ileum (IC50=0.82 pM), and enhances bradykinin (BK) induced contraction of isolated guinea pig ileum (EC50=0.383 pM)^[2].

In Vivo

CL-242817 (60 mg/kg, p.o., everyday for six weeks) causes male Wistar rats to fail to gain weight at a normal rate and significantly reduces cardiac function, significantly improves vascular occlusion and monocholinergic injury caused by Monocrotaline (HY-N0750), inhibits ACE activity and reduces hydroxyproline (HP) content in the lungs^[1].
