FAAH-IN-6 (compound 21d) is a potent, orally active and cross the blood-brain barrier fatty acid amide hydrolase (FAAH) inhibitor with IC₅₀s of 0.72, 0.28 nM for hFAAH, rFAAH, respectively. FAAH-IN-6 shows dose-dependent analgesic efficacy in animal models of both neuropathic and inflammatory pain.

In Vivo

FAAH-IN-6 (compound 21d) (1-10 mg/kg; p.o.) shows significantly ameliorates tactile allodynia in a dose-dependent fashion in SNI-induced neuropathic pain rats model^[1].
FAAH-IN-6 (3-10 mg/kg; p.o.) shows significantly ameliorates tactile allodynia of the ipsilateral hind paw in CFA-induced inflammatory pain model^[1].

For the following dissolution methods, please ensure to first prepare a clear stock solution using an In Vitro approach and then sequentially add co-solvents:

Protocol 1

Add each solvent one by one: 10% DMSO 90% Corn Oil

Solubility: 2.5 mg/mL (6.29 mM); Clear solution; Need ultrasonic

This protocol yields a clear solution of 2.5 mg/mL. If the continuous dosing period exceeds half a month, please choose this protocol carefully.

Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 900 μL Corn oil, and mix evenly.
