Jun13133 is a potent Enterovirus D68 3C Protease (EV-D68 3C^pro) inhibitor with an IC₅₀ of 28.6 nM. Jun13133 exhibits broad-spectrum and potent anti-enterovirus activity, including against EV-D68 (EC₅₀ = 8.2~9.9 nM) and EV-A71 (EC50 = 17.1 nM). Jun13133 covalently binds to the conserved catalytic residue Cys147 of EV-D68 3C^pro, thereby competitively inhibiting and blocking the ability of 3C^pro to catalytically cleave the viral polyprotein, thus inhibiting viral replication. Jun13133 can be used for research on severe respiratory disease and acute flaccid myelitis caused by EV-D68.

Jun13133 (50 mg/kg; i.p.; once daily; starting 24 h post-infection for 10 days) reduces paralysis and viral replication in EV-D68-infected neonatal mice^[1].
