MEISi-1

MEISi-1 is a small-molecule inhibitor targeting the homeodomain of the MEIS1 protein. MEISi-1 significantly inhibits the activity of luciferase reporter genes containing MEIS binding sites (TGACAG), and induces the self-renewal of a key type of mouse and human hematopoietic stem cells (HSC) both in vitro and in vivo. MEISi-1 promotes cardiomyocyte proliferation and cytokinesis, reduces cardiac fibroblast proliferation, and upregulates the expression of cardiomyocyte-specific genes. MEISi-1 can be used in research on hematological diseases, cardiac regeneration and cancer.

MEISi-1 (0.1-10 μM; 72 h) increases the proliferative capacity (up to 2.5-fold) and cytokinesis level (up to 2-fold) of neonatal rat ventricular cardiomyocytes[1].
MEISi-1 (5 μM; 72 h) reduces the viability/proliferative capacity of primary cardiac fibroblasts from adult mice[1].
MEISi-1 (10 nM-5 μM; 72 h) does not reduce the viability of human induced pluripotent stem cells (ATCC ACS-1026)[1].
MEISi-1 (5 μM; 18 days) upregulates pluripotency and cardiac mesoderm markers, increases the expression of the early cardiac marker Nkx2.5, decreases the expression of the late cardiac marker TNNT2, and inhibits the differentiation-associated upregulation of Meis1/2 expression in human induced pluripotent stem cells undergoing cardiomyocyte differentiation[1].
MEISi-1 (100 nM-5 μM; 3 days) upregulates the mesoderm marker Brachyury, the cardiac mesoderm marker GATA4, and the cardiac markers Nkx2.5 and TNNT2 during the cardiac mesoderm differentiation stage of human induced pluripotent stem cells, but does not alter the expression of ISL1[1].
MEISi-1 (0.1-10 μM; 7 days) significantly expands total hematopoietic cells from human umbilical cord blood and enriches CD34+, CD133+ and ALDHbr hematopoietic stem and progenitor cells in vitro[2].
MEISi-1 (0.1-10 μM; 7 days) significantly enriches multiple subsets of human mPB HSPCs in vitro, including the primitive CD34+CD38− cell population[2].
MEISi-1 (0.1-10 μM; 3 days) exerts no significant effect on the proliferation of human AD-MSCs or human HUVECs in vitro[2].
