Guide (The following is our recommended protocol. This protocol is only a guide and should be modified according to your specific needs).
1. Solution preparation
1.1 Preparation of stock solution
Solvent: H2O
Concentration: 50 mM is recommended.
Storage: Store at -20°C or -80°C in dark after aliquoting. Avoid repeated freezing and thawing.
1.2 Preparation of working solution
Dilute to 5 mM with cell culture medium (optimized according to the experiment).
Note: The working solution should be prepared and used immediately. Keep it away from light.
2. Cell staining
2.1 Cells are seeded in a 96-well plate at a concentration of 1.0-2.0?×?103 cells/well/100μL
2.2 Add 10 μL WST-8 at a final concentration of 5 mM.
2.3 Incubate cells at room temperature or 37°C for 1-4?h.
2.4 The absorbance at 450?nm was determined with a microplate reader.
