Tralkoxydim (PP604) is an orally effective herbicide. Tralkoxydim inhibits ACCase (IC50 of 0.5 μM in Lolium rigidum), blocking ATP-dependent acetyl-CoA carboxylation to malonyl-CoA and fatty acid biosynthesis; it activates HRP-catalyzed TMB/PDA peroxidation. Tralkoxydim can be used for research on hepatic porphyria.

In Vitro
Tralkoxydim (3-5 min) inhibits ACCase from both susceptible VLR 1 and resistant VLR 69 L. rigidum with an I50 of 0.5 µM, indicating no altered target-site resistance to tralkoxydim[1].
Tralkoxydim (1 μM; 15 min) inhibits maize (Zea mays L.) shoot ACCase in vitro, producing about 40% inhibition at 1 μM and concentration-dependent inhibition across the 125 nM-75 μM range, with about 25% inhibition at 125 nM and more than 80% inhibition at 75 μM[4].
Tralkoxydim (30 µM; 6 h) was metabolized 77% by resistant L. rigidum VLR 69 versus 41% by susceptible VLR 1, and this enhanced metabolism was not inhibited by PBO, malathion, ABT, or tetcyclacis, supporting a non-target-site resistance mechanism[1].
Tralkoxydim (1.0-5.0 mM) activates HRP-catalyzed TMB and PDA peroxidation in a concentration-dependent manner, increasing HRP activity to 203% for TMB and 212% for PDA at 5.0 mM, with mixed or noncompetitive activation depending on substrate and concentration[2].
Tralkoxydim (0-90 µg/mL) reduces Auxenochlorella sp. Z0 viability with an EC50 of 48.3 µg/mL[3].
Tralkoxydim addition correlates negatively with chlorophyll production in Z0 but positively in E11 and P19, with E11 and P19 showing higher chlorophyll and carotenoid contents under Tralkoxydim[3].
Tralkoxydim (noncytotoxic concentration; Day 7 postseeding) inhibits P-gp activity in MCF7R cells, with IC50 values of 88 μM using Rh123 accumulation and 90 μM using H33342 retention[7].

In Vivo
Tralkoxydim (100 mg/kg; oral gavage; 2 doses separated by 24 h) produces hepatic protoporphyrin accumulation in offspring of wild-caught Mus musculus, comparable to that in CD-1 mice[8].

- The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.
Add each solvent one by one: 10% DMSO 40% PEG300 5% Tween-80 45% Saline
Solubility: ≥ 5 mg/mL (15.18 mM); Clear solution
This protocol yields a clear solution of ≥ 5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (50.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
