Description

CNP7 is a covalent, selective HMGCS1 inhibitor. CNP7 reduces intracellular HMG-CoA levels. CNP7 induces global protein deisoprenylation. CNP7 exhibits a cell line-specific pattern of anticancer activity. CNP7 is applicable for cancer research[1].

In Vitro

CNP7 (2.5-20 μM; 72 h) exhibits cell line-specific antiproliferative activity across 18 cancer and non-cancer cell lines, with resistance correlated to higher baseline and CNP7-induced HMGCS1 expression in HCT116, HT29, and HepG2 cells, and sensitivity observed in RPE1, MFE296, and MOLM13 cells[1].

Cell Line: HCT116, HT29, HepG2, MCF7, PANC05.04, BT474, PC3, DU145, DLD1, Hela, U2OS, LnCap, RPE1, MFE296, MOLM13, JURKAT, MM.1S, SNU449
Concentration: 2.5-20 μM (cell viability assay); 1 μM (HMGCS1 expression analysis)
Incubation Time: 72 h (cell viability assay); 24 h (HMGCS1 expression analysis)
Result: Exhibited cell line-specific antiproliferative activity.
Showed high viability across all tested concentrations in resistant cell lines (HCT116, HT29, HepG2).
Caused significant dose-dependent viability reduction in sensitive cell lines (RPE1, MFE296, MOLM13).
Induced similar sensitivity curves that saturated at earlier doses in MM1.S and SNU449 cells (compared to simvastatin).
Demonstrated higher sensitivity in RPE1 and MFE296 cells than simvastatin.
Revealed higher baseline HMGCS1 expression in resistant cell lines, which was further elevated by treatment.
Showed lower baseline HMGCS1 expression and less pronounced increase upon treatment in sensitive cell lines.
