Biological Activity
Guide (The following is our recommended solution. It is for reference only. Please make modifications according to your specific needs.)
1. Preparation of stock solution[1] :
1) Dissolve CQ-Lyso in DMSO or DMF, and prepare a stock solution with a concentration of 1-10 mM.
2. Preparation of working solution:
Dilute the stored solution with pre-warmed serum-free cell culture medium or PBS to prepare a working solution of 1-10 μM.
3. Specific dyeing steps:
3.1 Cell Culture:
1) At 37°C, HeLa cells were cultured in DMEM medium containing 10% fetal bovine serum and 1% penicillin. When the adherent cells in the culture flask reached a confluence of 85%-95%, the cells were inoculated onto glass dishes and cultured for 24 hours. Before the experiment, the culture medium was discarded and the cells were washed three times with PBS buffer.
3.2 Staining Procedure:
Add 5.0 μM CQ-Lyso working solution to the above-processed cells and incubate at 37°C for 15 minutes.
2) (Optional) Multi-channel fluorescence imaging protocol: Add 1.0 μM of other dyes and continue incubation at 37°C for 15 minutes.
3. Fluorescence Imaging:
1) Fluorescence imaging was performed using a confocal microscope.
For CQ-Lyso, a 488 nm excitation light was used, and fluorescence signals were collected through the green fluorescence channel (515-550 nm) and the red fluorescence channel (570-620 nm, for clarity, the pseudo-color is yellow).
