Description

CRANAD-2 is a specific near-infrared (NIR) fluorescent probe for detecting plaques (unbound to Aβ: Ex=640 nm; Em=805 nm; after binding: Em=715 nm). CRANAD-2 penetrates the blood-brain barrier and has high affinity for Aβ aggregates, with a Kd value of 38 nM. CRANAD-2 is applicable to research related to Alzheimer's disease and neurodegenerative diseases[1][2][3][4].

In Vitro

CRANAD-2 (0-1500 nM; 1 h) binds to preformed ΔN amyloid fibrils with a dissociation constant of 55 ± 6 nM[1].
CRANAD-2 (35 nM) enables SMLM super-resolution imaging of preformed ΔN amyloid fibrils at a concentration of 35 nM, with a resolution of 45-55 nm[1].

In Vivo

CRANAD-2 (2.0 mg/kg; i.v.; single dose) enables in vivo detection of Aβ deposits in the arcAβ mouse model via MSOT and epi-fluorescence imaging, with specific co-localization to Aβ deposits in brain sections[2].

Animal Model: transgenic arcAβ mice (overexpressing human APP695 with Swedish and Arctic mutations, both sexes, 18-24 months-of-age); non-transgenic littermates (both sexes, 18-24 months-of-age)[2]
Dosage: 2.0 mg/kg
Administration: i.v.; single dose
Result: Exhibited higher cortical retention in arcAβ mice compared to non-transgenic littermates at 60-120 minutes post-injection; Co-localized with anti-Aβ antibodies in parenchymal and vessel-associated Aβ deposits in arcAβ mouse brain sections.
Showed no detectable signal in non-transgenic littermate cortex sections.
