Description

Cy5-DSPE chloride is a far-infrared fluorescent phospholipid (λex/λem = 600/665 nm). Cy5-DSPE chloride is used to label liposomes, membranes and lipid nanoparticles, and can be applied to tracking and imaging studies of liposomes[1].

In Vitro

Guide (The following is our recommended protocol. This protocol is only a guide and should be modified according to your specific needs).
1.\tStorage: Store powder at -20°C, protected from light.
2.\tDissolution: Dissolve Cy5-DSPE in chloroform, DMSO (HY-Y0320C), or ethanol to create a stock solution.
3.\tThin-Film Method:
3.1 Mix Cy5-DSPE stock solution with other lipids (e.g., PC, Cholesterol) in a round-bottom flask.
3.2 Evaporate the solvent (e.g., using a rotary evaporator) to form a thin lipid film.
3.3 Dry the film under vacuum to remove residual solvent.
3.4 Hydrate the film with buffer (e.g., PBS) and sonicate or extrude to form liposomes.
4.\tInsertion Method: Alternatively, add small amounts of Cy5-DSPE solution directly into pre-formed vesicle suspensions and incubate at a temperature near the lipid transition temperature to allow insertion into the membrane.
