Description

PREP-IN-3 is a mixed-type, selective prolyl endopeptidase (PREP) inhibitor. PREP-IN-3 can be used in research on neurological diseases^[1].

In Vitro

PREP-IN-3 (Compound Ce-3) (5 min-2 h) inhibits PREP in mouse brain S9 fraction with an IC₅₀ of 3.89 µM and acts as a mixed-type inhibitor with a K_i of 4.17 µM^[1].
PREP-IN-3 selectively inhibits PREP over DPP4, DPP8, DPP9, and FAP, with IC₅₀ values >100 µM for all four off-target enzymes^[1].
PREP-IN-3 (up to 10 µM; 4 h) in HT22 and SH-SY5Y cells, after treatment at concentrations up to 10 µM for 4 h, cell viability was >90%^[1].
PREP-IN-3 (2-10 µM) inhibited endogenous PREP activity in HT22 and SH-SY5Y neuronal cells in a concentration-dependent manner, with relative fluorescence decreasing to 64.12% at 2 µM and to 28.13-32.56% at 10 µM^[1].
