DMG-PEG2000-T7
DMG-PEG2000-T7 is a functionalized PEGylated lipid conjugate composed of three modular components: dimyristoylglycerol (DMG) as the lipid anchoring group, a polyethylene glycol (PEG) spacer arm with a molecular weight of 2000 Da, and T7 peptide (HAIYPRH) as the terminal targeting/functional ligand. DMG-PEG2000-T7 can be incorporated into liposomes, lipid nanoparticles (LNPs), or other nanocarriers to confer ligand-directed targeting capability against transferrin receptor (TfR/CD71) on the surface of cancer cells and brain endothelial cells.

Biological Activity
DMG-PEG2000-T7 is specifically designed for active targeting nanomedicine applications. It can be incorporated into liposomes, lipid nanoparticles (LNPs), or other nanocarriers to confer ligand-directed targeting capability against transferrin receptor (TfR/CD71) on the surface of cancer cells and brain endothelial cells.
Composition and role of each component:
1. DMG (dimyristoylglycerol): Lipid anchoring group-a diacylglycerol with two C14 chains, providing moderate membrane anchorage; its short-chain, high-fluidity characteristics enable faster dissociation of PEG-lipids from nanoparticle surfaces, facilitating endosomal escape and cellular uptake.
2. PEG2000 (polyethylene glycol, MW 2000): Hydrophilic polymer spacer arm that forms a steric corona around nanoparticles, reducing opsonization and reticuloendothelial system (RES) clearance, prolonging circulation half-life, and providing spatial separation between the lipid surface and targeting ligand.
3. T7 (T7 peptide (HAIYPRH)): Targeting ligand-a 7-amino acid peptide that binds to transferrin receptor (TfR/CD71), which is highly expressed on many rapidly proliferating cancer cells and brain capillary endothelial cells. Enables receptor-mediated transcytosis across the blood-brain barrier and targeted delivery to TfR-positive tumor cells as a non-immunogenic alternative to full-length transferrin. Target: transferrin receptor (TfR/CD71) on the surface of cancer cells and brain endothelial cells.
