In Vitro

DMG-pSar25 (50 ng firefly luciferase mRNA per well; 18 h) enhances mRNA delivery efficiency in ALC-0315 (HY-138300)-based LNPs in C2C12 and Hep3B cells compared to ALC-0159 PEG controls, while maintaining comparable cell viability[1].
DMG-pSar25 (50 ng firefly luciferase mRNA per well; 18 h) maintains mRNA delivery efficiency in SM-102 (HY-134541)-based LNPs in C2C12 and Hep3B cells compared to DMG-PEG2000 controls, while maintaining comparable cell viability[1].

In Vivo

DMG-pSar25 (0.06 µg mRNA; i.m.; single administration) replacing PEG lipids in ALC-0315 (HY-138170)-based LNPs enhances mRNA delivery efficiency (greater than 5-fold higher total flux) in healthy mice, while SM-102-based LNPs with DMG-pSar25 maintain comparable delivery efficiency to PEG-formulated LNPs, with similar immunogenicity profiles and no induction of anti-PEG antibodies[1].

Animal Model: C57BL/6J mice[1]
Dosage: 0.06 µg mRNA
Administration: i.m.; single administration
Result: Produced greater than 5-fold higher total flux (luminescence intensity) than ALC-0315-based LNPs formulated with ALC-0159 PEG lipid.
Resulted in significantly higher hEPO levels in muscle (P = 0.008) compared to ALC-0315-based LNPs with ALC-0159 PEG lipid; hEPO levels in blood were similar between the two groups.
Showed comparable total flux and hEPO levels (blood and muscle) to SM-102-based LNPs formulated with DMG-PEG2000.
Exhibited cytokine/chemokine profiles largely similar to PEG-formulated LNPs, with SM-102-based DMG-pSar25 LNPs showing significantly higher levels of G-CSF (P = 0.005), IL-6 (P = 0.0003), CXCL10 (P = 0.04), CXCL1 (P = 0.0008), M-CSF (P = 0.01), CXCL9 (P = 0.04), and CCL4 (P = 0.02) compared to SM-102-based DMG-PEG2000 LNPs; ALC-0315-based DMG-pSar25 LNPs showing significantly higher levels of IFNγ (P = 0.02), IL-6 (P = 0.008), IL-4 (P = 0.04), and IL-5 (P = 0.02), and significantly lower IL-2 (P = 0.04) compared to ALC-0315-based ALC-0159 LNPs.
Did not induce anti-PEG IgG production upon repeat administration, whereas repeat administration of PEG-formulated LNPs did (P = 0.04).
Did not cause significant elevation of plasma aspartate transaminase (AST) and alanine transaminase (ALT) levels compared to PBS-treated mice.
