DOPE-PEG2000-CTT2: surface modification of liposomes and lipid nanoparticles for tumor-targeted drug delivery and tumor imaging, with cyclic peptide structure providing enhanced stability and targeting affinity.
Structural composition:
(1) DOPE; a phospholipid with two unsaturated oleoyl chains that anchors into lipid bilayers and provides fusogenic activity for endosomal escape;
(2) PEG2000; a hydrophilic polymer that creates a steric barrier reducing opsonization and RES clearance, prolonging circulation time;
(3) CTT2 peptide; a cyclic tumor-targeting peptide that specifically binds to tumor vasculature or tumor cell surface markers, with the cyclic conformation (cyclized via disulfide or head-to-tail cyclization) providing enhanced proteolytic stability, improved binding affinity, and better selectivity compared to linear peptide counterparts, directing nanocarriers to tumor tissues for enhanced drug accumulation.
