DOPE-PEG2000-VIP

DOPE-PEG2000-VIP: surface functionalization of liposomes and nanoparticles for VPAC receptor-targeted drug delivery to tumors and for blood-brain barrier (BBB) crossing to deliver therapeutics to the central nervous system.
Structural composition:
(1) DOPE; a phospholipid with two unsaturated oleoyl chains that anchors into lipid bilayers and provides fusogenic activity for endosomal escape;
(2) PEG2000; a hydrophilic polymer that creates a steric barrier reducing opsonization and RES clearance, prolonging circulation time;
(3) VIP peptide; vasoactive intestinal peptide (28 amino acids) that binds to VPAC1 and VPAC2 G-protein-coupled receptors overexpressed on many tumor cells (breast, lung, prostate, colorectal cancers), enabling active tumor targeting; VIP also undergoes receptor-mediated transcytosis across the BBB, making it valuable for brain-targeted drug delivery in neurodegenerative diseases and brain tumors.
