DSG-PEG2000-iRGD
DSG-PEG2000-iRGD is a functionalized PEGylated lipid conjugate composed of three modular components: distearoylglycerol (DSG) as the lipid anchoring group, a polyethylene glycol (PEG) spacer arm with a molecular weight of 2000 Da, and iRGD peptide (HY-P0122) as the terminal targeting/functional ligand. DSG-PEG2000-iRGD can be incorporated into liposomes, lipid nanoparticles (LNPs), or other nanocarriers to confer ligand-directed targeting capability against sequential binding to αv integrins on tumor vasculature → neuropilin-1 (NRP1) in tumor tissue.

Biological Activity
DSG-PEG2000-iRGD is specifically designed for active targeting nanomedicine applications. It can be incorporated into liposomes, lipid nanoparticles (LNPs), or other nanocarriers to confer ligand-directed targeting capability against sequential binding to αv integrins on tumor vasculature → neuropilin-1 (NRP1) in tumor tissue.
Composition and role of each component:
1. DSG (distearoylglycerol): Lipid anchoring group-a diacylglycerol with two saturated C18 chains; provides strong membrane rigidity and sustained nanoparticle stability, suitable for applications requiring prolonged circulation time.
2. PEG2000 (polyethylene glycol, MW 2000): Hydrophilic polymer spacer arm that forms a steric corona around nanoparticles, reducing opsonization and reticuloendothelial system (RES) clearance, prolonging circulation half-life, and providing spatial separation between the lipid surface and targeting ligand.
3. iRGD (iRGD peptide (CRGDK/RGPD/EC)): Targeting ligand-a tumor-homing and penetrating peptide that combines an RGD motif (for αv integrin binding) with a C-end rule (CendR) sequence. Upon binding to αv integrins on tumor vasculature, the peptide is proteolytically processed to expose the CendR motif, triggering neuropilin-1 (NRP1)-mediated tissue penetration. This dual mechanism enables deep penetration into tumor tissue beyond the vasculature.
