Ferrichrome Iron-free

Ferrichrome Iron-free (Desferrichrome; DFC; N-Desferriferrichrome) is the ligand form of iron-loaded Ferrichrome when it is not bound to Fe3+. Ferrichrome Iron-free induces apoptosis in gastric cancer cells by activating the JNK-DDIT3 apoptotic axis. As an iron chelator, Ferrichrome Iron-free enables dual-modal imaging of HER2 via conjugation with SiR and Trastuzumab (HY-P9907). Ferrichrome Iron-free can be used in the research of gastric cancer and HER2-overexpressing cancers.

Activité biologique
Iron-free Ferrichrome (1-100 µg/mL; 72-120 h) inhibits the growth of MKN-7, MKN-45, MKN-74 and NUGC-4 gastric cancer cells in a dose-dependent manner, with the strongest inhibitory activity against MKN-7 (IC50 = 0.86 µg/mL) and MKN-45 (IC50 = 0.87 µg/mL) cells after 72 h of treatment[1].
Iron-free Ferrichrome (1-100 µg/mL; 72 h, 120 h, 3 h) induces the expression of apoptosis-related proteins (cleaved PARP, cleaved caspase-9, cleaved caspase-8) in MKN-7, MKN-45, MKN-74 and NUGC-4 gastric cancer cells, and upregulates the phosphorylation level of JNK in MKN-45 gastric cancer cells[1].
Iron-free Ferrichrome (1 µg/mL; 72 h) increases the population of apoptotic cells in the G1 subphase in MKN-74 gastric cancer cells, but does not alter the cell cycle distribution of MKN-7, MKN-45 or NUGC-4 gastric cancer cells[1].
Iron-free Ferrichrome (0.2-1 µg/mL; 48 h) dose-dependently induces DDIT3 mRNA expression in MKN-45 gastric cancer cells, and significantly induces DDIT3 mRNA expression in MKN-7 gastric cancer cells after 48 h of treatment, but does not alter DDIT3 expression in MKN-74 or NUGC-4 gastric cancer cells[1].
Iron-free Ferrichrome (1-100 µg/mL; 24 h) reduces intracellular iron levels in MKN-7, MKN-45, MKN-74 and NUGC-4 gastric cancer cells to similar levels[1].
