Ferroptosis inducer-12 is a potent and selective ferroptosis inducer. Ferroptosis inducer-12 exhibits potent antiproliferative activity against HT1080 and OS-RC-2, with IC50 values of 3 nM and 5 nM, respectively. Ferroptosis inducer-12 strongly inhibits GPX4 enzymatic activity, induces intracellular ROS and elevates intracellular Fe2+ levels in OS-RC-2 cells. Ferroptosis inducer-12 significantly inhibits tumor growth in BALB/c nude mice bearing OS-RC-2 xenografts. Ferroptosis inducer-12 can be used for the study of cancer.

Biological Activity
Ferroptosis inducer-12 (Compound A6) (48 h) exhibits potent antiproliferative activity against HT1080 and OS-RC-2, with IC50 values of 3 nM (HT1080) and 5 nM (OS-RC-2)[1].
Ferroptosis inducer-12 (1 μM, 2 h) strongly inhibits glutathione peroxidase 4 (GPX4) enzymatic activity in OS-RC-2 cells[1].
Ferroptosis inducer-12 (0.5 μM, 4 h) induces intracellular ROS, elevates intracellular Fe2+ levels and promotes lipid peroxidation (LPO) accumulation in OS-RC-2 cells[1].
Ferroptosis inducer-12 (10 nM, 24 h) significantly suppresses migration and reduces invasive capacity of OS-RC-2 cells[1].

Animal Model:OS-RC-2 cells (2 × 106 in 100 μL PBS) were subcutaneously implanted into the right limb subcutaneous fat of 4-week-old male BALB/c nude mice (weight: 14-16 g)[1]

Result:Showed tumor growth inhibition rate of 44.59% at 50 mg/kg, and reduced tumor volume and weight.

- To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for In Vivo experiments, it is recommended to prepare freshly and use it on the same day.
- The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.
Add each solvent one by one: 10% DMSO 40% PEG300 5% Tween-80 45% Saline
Solubility: ≥ 2.5 mg/mL (4.17 mM); Clear solution
This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
