Fura-2 AM (solution) (Fura-2 Acetoxymethyl ester (solution)) is a membrane permeable, intracellular, UV light-excitable and ratiometric fluorescent Ca2+ (calcium ion) indicator. Fura-2 AM crosses cell membranes and is converted to Fura-2 (HY-D0110A) via cellular esterases. Fura-2 AM can be used to detect calcium in cells.

Biological Activity

Guide (The following is our recommended protocol. This protocol only provides a guideline, and should be modified according to your specific needs).
Fura-2 AM diffuses across the cell membrane and is de-esterified by intracellular esterases to generate free Fura-2 acid.
1. Aliquot 2 mL of culture medium into a conical tube and warm it to 37°C. Add 2 μL of Fura-2 AM stock solution to prepare a 1 μM Fura-2 AM solution. Vortex vigorously for 1 min.
2. Transfer the loading solution into a tissue culture dish, and move the cell-containing coverslip into the dish.
3. Incubate neurons at 37°C in a dark incubator for 30 min. Accurately record the incubation time.
4. Prepare a new culture dish containing 2 mL tissue culture medium without Fura-2 AM. Remove the coverslip from the loading solution and transfer it into the new dish.
5. Mount the coverslip onto the imaging chamber.
