C14-306
C14-306 is an ionizable lipid with a linear 306 amine core and a C14 alkyl tail. C14-306 enhances brain transfection efficiency. Lipid nanoparticles (LNP) formulated with C14-306 promote mRNA delivery, increase neuronal transfection rate, and facilitate Cre-mediated recombination in the brain, while reducing off-target expression in the liver.

Actividad biológica
LNPs formulated with C14-306 (60 ng mRNA/20,000 cells; 24 h) efficiently and specifically transfect the neuronal cell line SH-SY5Y in vitro, exhibit extremely low transfection activity toward the brain microvascular endothelial cell line hCMEC/D3, and induce high uptake by neuronal cells[1].
MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.
LNPs formulated with C14-306 (0.35 mg/kg encapsulated mRNA; i.v.; single dose), particularly LNP 14, drive potent, liver-sparing mRNA transfection in the healthy Mus musculus brain, with efficacy far exceeding FDA-approved and benchmark LNPs[1].
The C14-306-containing LNP 14 (0.35 mg/kg encapsulated mRNA; i.v.; single dose) does not induce BBB leakage, significant systemic inflammation, or liver enzyme elevation in healthy mice, demonstrating a favorable safety profile[1].
