NVS-STG2 is a molecular glue that targets STINGSTING by binding to pockets between adjacent STING dimer transmembrane domains, effectively acting as a molecular glue. NVS-STGI enhances the activity of cGAMP by inducing the formation of more abundant and larger oligomers. NVS-STG2 produces antitumor activity in animal models.

NVS-STG2 (50 μM; 24 h) strongly induces ISRE-Luc reporter gene expression in a reporter gene assay in THP1-Dual cells, AC50=5.2 μM[1].
NVS-STG2 (50 μM; 16 h) induces STING-dependent IRF3 phosphorylation in a STING co-transfection reporter gene assay in HEK293T cells[1].
NVS-STG2 (50 μM; 16 h) causes wild-type STING to form bright punctate structures in a STING translocation/aggregation assay in 293T cells, but not the R95A mutant[1].
NVS-STG2 (40 μM; 16 h) induces the formation of higher-order oligomers of hSTING in native gel analysis of purified hSTING protein[1].

This protocol yields a clear solution of 2.5 mg/mL.
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
Add each solvent one by one: 10% DMSO 90% (20% SBE-β-CD in Saline)
Solubility: 2.5 mg/mL (5.85 mM); Clear solution; Need ultrasonic
This protocol yields a clear solution of 2.5 mg/mL.
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 900 μL 20% SBE-β-CD in Saline, and mix evenly.
Preparation of 20% SBE-β-CD in Saline (4°C, storage for one week): 2 g SBE-β-CD powder is dissolved in 10 mL Saline, completely dissolve until clear.
