Description

Green DND-26 is a green fluorescently labeled lysosomal probe with a maximum excitation/emission wavelength of 504/511 nm. The structure is composed of a fluorescein group and linked weak bases, which can freely cross the cell membrane and generally gather on spherical organelles. Green DND-26 is suitable for observing the internal biosynthesis and related pathogenesis of lysosomes[1].

In Vitro

Guide (The following is our recommended protocol. This protocol is only a guide and should be modified according to your specific needs).
1. Preparation of Stock Solution and Working Solution
1.1 Prepare a 1 mM stock solution using anhydrous DMSO.
1.2 Dilute the stock solution with preheated serum-free cell culture medium or PBS to prepare a 50-100 nM working solution.
Note: Please adjust the concentration of the dye working solution according to the actual situation, and prepare it fresh before use.
2. Cell Staining
2.1 Suspension Cells: Centrifuge at 1000 g for 3-5 minutes at 4°C, discard the supernatant. Wash twice with PBS, 5 minutes each time.
Adherent cells: Discard the cell culture medium, wash twice with PBS, 5 minutes each time.
2.2 Add Green DND-26 working solution and incubate at room temperature in the dark for 30 minutes.
2.3 Discard the staining solution, wash twice with PBS, 5 minutes each time.
2.5 Resuspend the cells in serum-free cell culture medium or PBS, and analyze using a fluorescence microscope or flow cytometry.
