Product Information
Heparinase has a theoretical molecular weight of 42.5 kDa and an isoelectric point of 9.3-9.5. It can selectively (by an elimination mechanism) cleave highly sulfated polysaccharide chains containing 1-4 linkages between hexosamine and O-sulfated iduronic acid residues. The reaction produces oligosaccharide products (mainly disaccharides) containing unsaturated uronic acids, which can be detected by UV spectroscopy at 232 nm.
Dissolution
The enzyme is typically dissolved in 20 mM Tris-HCl (pH 7.5), 50 mM NaCl, 4 mM CaCl2 and 0.01% BSA at a concentration of 1 mg/mL.
Reaction conditions
1. pH stability range 4.0-9.0, optimum pH 7.0-7.5
2. Temperature stability range 20-37°C, optimum temperature 30°C
3. Heparinase is calcium-dependent and has maximum activity under 1-5 mM CaCl2 conditions. It is recommended to add CaCl2 to the reaction system.
Note: The incubation time and enzyme concentration of the enzyme-cleaved substrate need to be determined based on actual experience.
