AS15
AS15 is a PDIA1 inhibitor with an IC50 value of 300 nM. AS15 interacts with the active-site cysteine of PDIA1, and this process is independent of the substrate-binding domain of PDIA1. AS15 acts as a synergistic cell growth inhibitor with buthionine sulfoximine (BSO) (HY-106376), a glutathione synthesis inhibitor. AS15 can be used in the research of glioblastoma.

Biological Activity
|
PDIA1 300 nM (IC50) |
AS15 reduces the viability of U-87 MG glioblastoma cells, with an IC50 of 18.3 μM[1].
AS15 (0.3-3 μM; 24 h) reduces the colony-forming ability of U-118 MG glioblastoma cells, and its activity can be enhanced by glutathione depletion via pre-treatment with BSO[1].
MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.
-
Cell Line:U-118 MG glioblastoma cells
-
Concentration:0.3, 1 and 3 μM
-
Incubation Time:24 h (BSO pretreatment)
-
Result:Increased potency in reducing colony formation after pretreatment with BSO (a glutathione synthesis inhibitor), supporting that glutathione depletion sensitizes glioblastoma cells to AS15.
