QTZ is a bioluminescence agent for in vivo imaging. QTZ has red-shifted emission and yields very little background. QTZ is a coelenterazine analog with the 4-quinolinyl substitution at the C8 position of the imidazopyrazinone core.

Biological Activity
Guide (The following is our recommended experimental protocol. This protocol is for reference only and should be adjusted according to your specific needs).
1. Solution preparation[1]
1.1 Preparation of stock solution
Solvent: DMSO
Concentration: 25 mM (optimized according to the experiment).
Storage: Store at -20°C or -80°C in dark after aliquoting. Avoid repeated freezing and thawing.
1.2 Preparation of working solution
Dilute to 25-50 μM with DPBS or other buffers.
Note: The working solution should be prepared and used immediately. Keep it away from light.
2. Cell staining
HeLa and MDA-MB-453 cells, and dilute to 1 × 105 cells/mL with DPBS.
10 μg of purified scFV-QLuc fusion protein was added to 1 mL of cell suspension and incubated on ice for 30 min.
Cells were pelleted by centrifugation and resuspended in 1 mL of DPBS.
100 μL of cell suspension was added to a 96-well plate.
100 μL of 50 μM QTZ in DPBS was added to the cell suspension to a final concentration of 25 μM.
Bioluminescence intensity was recorded using a microplate reader equipped with an extended range red-sensitive photomultiplier tube.

Anesthetize the Her2+ tumor.
The scFv-QLuc fusion protein (1 mg) diluted with normal saline to a volume of 100 μL was administered through the tail vein.
6 hours later, intravenously inject 100 μL QTZ (1 mM) (25% (w/v) HP-β-CD and 20% PEG-400 dissolved in normal saline).
Bioluminescence imaging is performed immediately after substrate administration in the aforementioned Bioluminescence imaging dark box.
