Pifexole (5-20 mg/kg; i.v.; single administration; monitored for approximately 20-25 min) inhibits the polysynaptic crossed extensor reflex in cats anesthetized with chloralose and urethane, with inhibition rates of 81.6% and 93.1% at 5 mg/kg and 10 mg/kg, respectively, but does not affect the monosynaptic patellar tendon reflex at 10-20 mg/kg^[2].
Pifexole (15.4-200 mg/kg; p.o.) inhibits electroshock-induced convulsions in rats and mice with ED₅₀ values of 17.5 mg/kg and 15.4 mg/kg, respectively, and inhibits strychnine-induced convulsions in both species with an ED₅₀ of 96 mg/kg, while antagonizing nicotine toxicity in mice with an ED₅₀ of 30 mg/kg. Pifexole also reduces muscle strength in mice by 25% and 35% at 100 mg/kg and 200 mg/kg, respectively, inhibits spontaneous activity in mice at 100 mg/kg, and reduces morphine-induced excitation and hyperactivity at 200 mg/kg^[2]
