Jun13133
Jun13133 is a potent Enterovirus D68 3C Protease (EV-D68 3Cpro) inhibitor with an IC50 of 28.6 nM. Jun13133 exhibits broad-spectrum and potent anti-enterovirus activity, including against EV-D68 (EC50 = 8.2~9.9 nM) and EV-A71 (EC50 = 17.1 nM). Jun13133 covalently binds to the conserved catalytic residue Cys147 of EV-D68 3Cpro, thereby competitively inhibiting and blocking the ability of 3Cpro to catalytically cleave the viral polyprotein, thus inhibiting viral replication. Jun13133 can be used for research on severe respiratory disease and acute flaccid myelitis caused by EV-D68.

Result:Ameliorated paralysis induced by viral infection, with paralysis scores significantly lower than those of the vehicle control group.
Prevented excessive weight loss in mice (weights were significantly higher on days 12-14 post-infection compared to the control group), indicating reduced disease severity.
Reduced viral titers in the spinal cord at 4 and 7 days post-infection (virus was almost undetectable).
Decreased viral titers in the injected quadriceps (infected muscle tissue) at 4 and 7 days post-infection.
