Ac4GlcNAcF3
Ac4GlcNAcF3 is a cell-permeable glycomimetic that can be recognized and utilized by OGT and is resistant to hydrolysis by OGA. GlcNAcF3, the active metabolite of Ac4GlcNAcF3, can be recognized by OGT as a substrate for peptide modification. Ac4GlcNAcF3 upregulates the level of cellular O-GlcNAc modification. Ac4GlcNAcF3 can be used in studies related to glycosylation regulation.

Biological Activity
Ac4GlcNAcF3 produces GlcNAcF3 as its active metabolite, which is recognized by OGT as a substrate for peptide modification in peptide-based in vitro assays but cannot be cleaved by OGA[1].
Ac4GlcNAcF3 (0-100 μM; 0-36 h) significantly increases the level of O-GlcNAcylation in NIH3T3 cells[1].
Ac4GlcNAcF3 (20 μM; 24 h) exerts an OGT expression-dependent O-GlcNAcylation-enhancing effect in NIH3T3 cells, as reduced OGT levels significantly attenuate this effect[1].
Ac4GlcNAcF3 (0-100 μM; 48 h) exhibits no significant cytotoxicity against NIH3T3, MCF7, HepG2 or HeLa cells[1].
Ac4GlcNAcF3 (20 μM; 24 h) increases the level of O-GlcNAc glycosylation in NIH3T3 cells[1].
