DMG-PEG2000-TAT

DMG-PEG2000-TAT is specifically designed for active targeting nanomedicine applications. It can be incorporated into liposomes, lipid nanoparticles (LNPs), or other nanocarriers to confer ligand-directed targeting capability against non-specific cell membrane penetration (heparan sulfate proteoglycans).
Composition and role of each component:
1. DMG (dimyristoylglycerol): Lipid anchoring group-a diacylglycerol with two C14 chains, providing moderate membrane anchorage; its short-chain, high-fluidity characteristics enable faster dissociation of PEG-lipids from nanoparticle surfaces, facilitating endosomal escape and cellular uptake.
2. PEG (polyethylene glycol): Hydrophilic polymer spacer arm that forms a steric corona around nanoparticles, reducing opsonization and reticuloendothelial system (RES) clearance, prolonging circulation half-life, and providing spatial separation between the lipid surface and targeting ligand.
3. TAT (TAT peptide (YGRKKRRQRRR): Targeting ligand-a classical cell-penetrating peptide (CPP) derived from the transactivation domain of HIV-1 Tat protein. Its highly cationic arginine-rich sequence enables efficient transmembrane transport through multiple uptake mechanisms (macropinocytosis, direct penetration). It lacks cell-type specificity but provides potent non-specific cellular uptake enhancement.
