DOPE-PEG2000-R8

DOPE-PEG2000-R8: surface modification of liposomes and nanoparticles for enhanced intracellular delivery of various cargoes including small-molecule drugs, nucleic acids (DNA, siRNA), peptides, and imaging agents.
Structural composition:
(1) DOPE; a fusogenic phospholipid with two oleoyl chains that anchors into lipid bilayers and promotes membrane destabilization and fusion for endosomal escape;
(2) PEG2000; a hydrophilic polymer that provides steric stabilization and prolongs circulation half-life by reducing immune clearance;
(3) R8 peptide; octaarginine, a highly cationic cell-penetrating peptide (CPP) with eight arginine residues that facilitates rapid cellular uptake through electrostatic interactions with negatively charged cell membrane components (heparan sulfate proteoglycans, phospholipids), enabling translocation of nanocarriers across cell membranes via multiple mechanisms including macropinocytosis and direct membrane penetration.
