Monoamine oxidase (EC 1.4.3.4) is an enzyme composed of different polypeptides. Monoamine oxidation catalyzes the oxidative deamination of various biological amines in brain and peripheral tissues by producing hydrogen peroxide. Monoamine oxidase plays an important role in maintaining the regulation of synaptic transmission, emotional behavior and other brain functions.

Actividad biológica
Monoamine oxidase has some isoenzymes, the most typical of which are monoamine oxidase (MAO) A and MAOB. MAOA is usually inhibited by low concentrations of Clorgyline (HY-14197A) and MAOB is usually inhibited by low concentrations of l-deprenyl[3].
Monoamine Oxidase Assay
1. Solution Preparation
1) Substrate: 1.0% v/v (0.10 M) benzylamine in 67 mM potassium phosphate buffer, pH 7.2.
2) Enzyme solution: Dissolve to a concentration of 10 mg/mL in 67 mM potassium phosphate buffer (pH 7.2) and store at 2-8°C for up to 1 week.
2. Assay Procedure
1) Adjust spectrophotometer to 250 nm and 25°C.
2) Pipette 2.8 mL 67 mM Potassium phosphate buffer pH 7.2 and 0.1 mL substrate solution into each cuvette.
3) Incubate in spectrophotometer for 3-4 minutes to reach temperature equilibrium.
4) Add 0.1 mL enzyme solution and record the increase in A250 over 6-8 minutes. Calculate ΔA250/min from the linear portion of the curve. A 2-3 minute delay may occur.
5) Calculation
Units/g = (ΔA250/min x 1000)/(13.0 x mg enzyme/mL reaction mixture)
