N-Methylmesoporphyrin IX (NMM) (solution) , a widely used G-quadruplex DNA specific fluorescent binder, is an efficient probe for monitoring Aβ fibrillation. N-Methylmesoporphyrin IX is an in situ inhibitor and an ex situ monitor for Aβ amyloidogenesis both in vitro and in cells. N-Methylmesoporphyrin IX is sensitive to G-quadruplexes DNA but has no response to duplexes, triplexes and single-stranded forms DNA. N-Methylmesoporphyrin IX is nonfluorescent alone or in monomeric Aβ environments, but emits strong fluorescence through stacking with the Aβ assemblies.

Activité biologique
Solvent and concentration: DMSO: 2 mM
Guide (Following is our recommended protocol. This protocol only provides a guideline, and should be modified according to your specific needs).
1. Preparation of solution preparation
Dissolve N-Methylmesoporphyrin IX in DMSO (5-10 mg/mL) or 0.2 M HCl.
2. G-Quadruplex Staining Protocol (Gel):
2.1 Fold DNA in a buffer containing KCl (e.g., 50 mM KCl).
2.2 Run electrophoresis on a 12-15% polyacrylamide gel.
2.3 Stain the gel with 10 μg/mL N-Methylmesoporphyrin IX in 1x G4 folding buffer for 15 min under agitation.
2.4 Keep the gel in the dark to avoid losing signal.
3. Solution Titration/Fluorescence Study:
3.1 Prepare N-Methylmesoporphyrin IX in working buffer (e.g., 10 mM lithium cacodylate, pH 7.2, 5 mM KCl, 95 mM LiCl).
3.2 Co-incubate N-Methylmesoporphyrin IX (e.g., 1 μM) with DNA (e.g., 50 μM).
3.3 Monitor fluorescence, ensuring proper KCl concentration for G4 folding.
