NRX-103094 is a Molecular glue and β-catenin/β-TrCP interaction enhancer, with an EC50 of 62 nM for the binding of pSer33/S37A β-catenin peptide to recombinant β-TrCP/Skp1 complex. NRX-103094 shows selectivity for Ser37 phosphorylation-deficient β-catenin, and has no effect on wild-type doubly phosphorylated β-catenin or other β-TrCP substrates. NRX-103094 can be used in research related to colorectal cancer and ovarian endometrioid adenocarcinoma.

NRX-103094 acts as a canonical molecular glue, enhancing the binding affinity between β-catenin degron peptide and β-TrCP E3 ligase without detectable individual affinity for either protein partner[1].
NRX-103094 enhances the binding of pSer33/Ser37 β-catenin peptide to recombinant β-TrCP/Skp1 complex in a TR-FRET assay with an EC50 of 457 nM[2].
NRX-103094 enhances the binding of pSer33/S37A β-catenin peptide to recombinant β-TrCP/Skp1 complex in a TR-FRET assay with an EC50 of 62 nM and 1000-fold binding cooperativity at 40 μM[2].
NRX-103094 (0.006-25 μM) dose-dependently potentiates ubiquitylation of pSer33/S37A β-catenin peptide by recombinant SCFβ-TrCP, achieving levels exceeding WT β-catenin peptide at 250 nM[2].
