Biological Activity
Guide (The following is our recommended protocol. This protocol is only a guide and should be modified according to your specific needs).
Particle Preparation[1]
1. Dilute 250 μL of a 5.75% polystyrene particle suspension with 0.1 M sodium bicarbonate buffer (pH 8.3) to a total volume of 1.5 mL, achieving a 6-fold dilution. Then, place the mixture in an ice bath and sonicate for 5 minutes.
2. While stirring gently, slowly add 1.5 mL of 300 μM OG 488, SE labeling reagent and an equal amount of 0.1 M sodium bicarbonate buffer (pH 8.3) to the solution.
3. After gently stirring, let the mixture incubate in a dark environment at room temperature for 2 hours.
4. Next, take 15 μL of 40 μM Texas Red and SE labeling reagent dissolved in dimethylformamide and carefully add it to the particle solution under ultrasonic treatment to ensure even dispersion.
5. After incubation, slowly centrifuge the particles at 1000 g for 10 minutes, collect the pellet, and wash it three times with 0.1 M sodium bicarbonate buffer (pH 8.3), finally resuspending the particles in the same buffer for storage. The polystyrene particles are labeled with fluorescent dyes using a chemical labeling method for subsequent experimental applications.
