Description

PROTAC stands for proteolysis-targeting chimeras, which are heterobifunctional small-molecule compounds containing two different ligands: one is a ubiquitin ligase E3 ligand, and the other is a ligand that binds to the target protein in cells. The two ligands are connected by a linker, thereby forming a "ternary" polymer-target protein ligand-Linker-E3 ligand. PROTACs bring the target protein and the intracellular E3 ubiquitin ligase into close proximity, utilizing the "ubiquitin-proteasome" pathway to specifically degrade the target protein. PROTACs can improve the druggability of "undruggable" targets.
