Biological Activity
pNPPT (compound 2a) cyclohexanamine is efficiently hydrolyzed by wild-type PP1γ, with a maximum catalytic efficiency of 31 M-1s-1 and a second-order rate acceleration ratio of 1.7 × 1011. The transition state is characterized by restricted leaving group bond cleavage and effective general acid neutralization of the leaving group. The hydrolysis catalytic efficiency decreases for the PP1 R96K and PP1 R221K single mutants of PP1γ, with corresponding catalytic efficiency values of 0.8 M-1s-1 and 1.9 M-1s-1[1].
