PSMA-SulfoCy7 is a high-affinity imaging agent targeting PSMA/GCPII (with a Ki of 18.1 nM for human PSMA). PSMA-SulfoCy7 regulates PSMA-dependent NAAG hydrolysis. PSMA-SulfoCy7 exhibits excellent in vivo imaging capability, enabling clear visualization of PSMA-expressing tumors in xenograft models, with no obvious toxicity even at a dose of 87.9 mg/kg. PSMA-SulfoCy7 is widely used in prostate cancer-related studies[1].

In Vitro

PSMA-SulfoCy7 (2 h; 60 min) inhibits PSMA-dependent NAAG hydrolysis in LNCaP cell lysate with a Ki of 18 nM[1].
PSMA-SulfoCy7 is cytotoxic to LNCaP and PC-3 prostate cancer cell lines with CC50 values of 12 μM and 19 μM, respectively[1].

PSMA-SulfoCy7 (250 nmol/kg; i.v.; single dose) demonstrates specific accumulation in PSMA-positive 22Rv1 prostate cancer xenografts, with a twofold higher in vivo fluorescence signal than PSMA-negative PC-3 xenografts at 24 hours post-single intravenous administration of 250 nmol/kg[1].
PSMA-SulfoCy7 (i.v.; single dose) exhibits biphasic elimination from blood plasma in rabbits and rats, with elimination half-lives of 1.39 h and 1.78 h, respectively, and primarily distributes to tissues with elimination via the urinary system[1].
