FGT-4 is a FR-β-targeted chimeric molecule. FGT-4 acts as a TLR7 agonist. FGT-4 binds to FR-β to enable delivery to tumor-associated macrophages and myeloid-derived suppressor cells. FGT-4 promotes the secretion of iNOS (associated with M1 macrophages) and the pro-inflammatory cytokine IL-6, and enhances the proliferation of cytotoxic CD8+ T cells. FGT-4 can be used for the research of breast cancer .

Poly (U) (Polyuridylic acid) is an RNA chain that exists in a disordered extended state at room temperature and serves as a TLR7 ligand. Poly (U) produces distinguishable blocking effects when transported through α-hemolysin nanopores. Poly (U) forms a triple helix with adenosine or adenosine monophosphate, thereby significantly promoting the nucleotide bond condensation reaction between adenosine monophosphate and adenosine, while exerting minimal effects on other nucleosides. When conjugated with cationic lipids, Poly (U) specifically stimulates plasmacytoid dendritic cells to produce type I interferons .

Lipid N2-3L is an ionizable cationic lipid (pKa = 8.99) that can be used to generate supramolecular lipid nanoparticles (SMLNPs) for mRNA delivery. Lipid nanoparticles formed by encapsulating a luciferase reporter gene with Lipid N2-3L were detected to accumulate in the lymph nodes of mice, indicating that they can effectively enter the mouse immune system. Lipid nanoparticles formed by encapsulating ovalbumin mRNA and the TLR7/8 agonist Resiquimod (HY-13740) with Lipid N2-3L were able to reduce tumor volume and increase survival in the MC-38-OVA mouse colon cancer model. Lipid N2-3L can be used in the research of drug delivery for cancer-related therapies .

ENDO12 is an inhibitor of the Munc13-4-STX7 protein complex, with a Kd value of 2.7 µM for STX7. ENDO12 blocks the interaction of Munc13-4-STX7. ENDO12 inhibits endolysosomal flux, endolysosomal cargo degradation, the extracellular signal-regulated kinase signaling pathway in neutrophils, the IFN regulatory factor signaling pathway in plasmacytoid dendritic cells, and the responses of primary dendritic cells to TLR3, TLR7, and TLR9. ENDO12 alleviates CpG-induced systemic inflammation by reducing the levels of myeloperoxidase, IL-6 and IFNγ. ENDO12 does not interfere with the host's antiviral response to lymphocytic choriomeningitis virus infection.\nENDO12 can be used in studies related to systemic inflammation .
