AZM198 is an orally active myeloperoxidase (MPO) inhibitor. AZM198 irreversibly inactivates MPO (IC50=0.015 μM) via covalent binding to the heme prosthetic group, preferentially targets extracellular MPO activity, and reduces neutrophil extracellular trap formation, reactive oxygen species production and degranulation. AZM198 increases the fibrous cap thickness of atherosclerotic plaques, reduces lesion area, ameliorates hepatic steatosis and fibrosis in non-alcoholic steatohepatitis, and alleviates proteinuria and inflammatory infiltration associated with glomerulonephritis. AZM198 also decreases circulating levels of high-sensitivity Cardiac Troponin I and IL-1β, and mitigates endothelial cell injury. SL-017 is a novel photoacoustic sensitizer and a derivative of photofrin B. It can be taken up by cells to the maximum extent within 30 minutes and is mainly localized in mitochondria. After being activated by visible light or ultrasound, SL-017 can significantly increase the production of reactive oxygen species (ROS). Low concentrations of SL-017 can rapidly cause the loss of mitochondrial membrane potential. SL-017 can also cause mitochondrial fragmentation, a process that occurs after the loss of membrane potential. Epoxyeicosatrienoic acids (EETs) can alleviate the loss of mitochondrial membrane potential caused by SL-017, but the antioxidant ascorbic acid has no such effect. These characteristics indicate that SL-017 mainly targets mitochondria and exerts its cytotoxic effect by triggering the collapse of mitochondrial membrane potential, generating ROS, and causing mitochondrial fragmentation. As a novel photoacoustic sensitizer, SL-017 has potential application value in photodynamic therapy and sonodynamic therapy.

NSC 109555 is an ATP-competitive inhibitor of checkpoint kinase 2 (Chk2; IC50=200 nM in a cell-free kinase assay). LHA585 is a fluorescent probe targeting the lipid hydrogen abstraction process for the early detection of ferroptosis (Ferroptosis). Its detection mechanism relies on the reactivity of its own 4-phenyl-3-methylbut-2-enyl moiety with biologically relevant oxidative radicals, including peroxyl radicals (ROO·), alkoxyl radicals (RO·), hydroxyl radicals (HO·), and peroxynitrite anions (ONOO−); upon hydrogen abstraction from this structure, radical rearrangement and radical cleavage occur, releasing a xanthene radical which converts into a highly fluorescent rhodamine dye in biological environments, thereby achieving a fluorescence turn-on ratio of up to 360-fold.
