Stellettamide A TFA is a marine toxin found in a marine sponge. Stellettamide A TFA is a calmodulin inhibitor. Stellettamide A TFA can inhibit Ca2+/Mg2+ ATPase, phosphodiesterase, myosin light chain , and Mg2+-ATPase. Stellettamide A TFA inhibits high K+- and Ca2+-induced contraction in permeabilized smooth muscle. Stellettamide A TFA exhibits antifungal activity against Mortierella remannianus. Stellettamide A TFA can be used for the research of fungal infection[1].

In Vitro

Stellettamide A TFA (3-300 μM; 1 h) inhibits Ca2+-stimulated Mg2+-ATPase activity in native actomyosin from chicken gizzard with an IC50 of 25 μM[1].
Stellettamide A TFA (3-300 μM) inhibits myosin light chain phosphorylation in a reconstituted smooth muscle contractile system with an IC50 of 152 μM, and this effect is antagonized by higher calmodulin concentrations[1].
Stellettamide A TFA (3-300 μM) inhibits Ca2+/calmodulin-dependent (Ca2+-Mg2+)-ATPase from rabbit erythrocyte membranes (IC50 = 100 μM) and phosphodiesterase from bovine heart (IC50 = 52 μM), with the latter effect antagonized by higher calmodulin concentrations[1].
Stellettamide A TFA (3-300 μM) quenches calmodulin’s intrinsic tyrosine fluorescence in a concentration-dependent manner with or without Ca2+, indicating direct binding independent of Ca2+[1].
Stellettamide A TFA inhibits high
K+(72.7 mM), Ca2+(3 mM and 10 mM)-induced contraction in the smooth muscle of guinea-pig taenia coli with an IC50 of 88, 46 and 105 mM[1].
