TSQ (solution)
TSQ (solution) is a cytosolic zinc ion fluorescence probe that is membrane permeable and can be used for intracellular imaging of zinc proteins (λmax ~470 nm). TSQ can combine with Zn2+ in the presence of Ca2+ and Mg2+ to produce blue fluorescence (Ex/Em = 334/495 nm) .

Guidelines (Following is our recommended protocol. This protocol only provides a guideline, and should be modified according to your specific needs).
1. Solution preparation[4]
1.1 Stock solution
Storage: Store at -20°C or -80°C in dark after aliquoting. Avoid repeated freezing and thawing.
1.2 Preparation of working solution
Dilute with DPBS or other buffers. The corresponding stock solution can be diluted according to the actual situation. Note that if the solvent is DMSO, the cytotoxicity of DMSO must be considered, and a solvent control should be prepared; if the solvent is pure water, the working solution needs to be filtered and sterilized before adding cells.
Note: The working solution should be prepared and used immediately. Keep it away from light.
2. Cell staining
2.1 Cells are grown on glass coverslips coated with Collagen, rat tail (HY-NP010).
2.2 Rinsed three times with Dulbecco's phosphate buffered saline.
2.3 30 μM TSQ in DPBS, mix well and incubate at 37°C for 30 minutes.
2.4 After the stained cells are rinsed three times with DPBS.
2.5 Fluorescence microscopy detection (Ex/Em = 334/495 nm).
