Intracellular Zn2+ ions detection by TSQ (0.1 M; 30 min). The results showed that the intracellular Zn2+ ion content in the nanoparticle groups was significantly higher than that in the control group, with the siRNAPD−L1@HA-ZIF-8 group exhibiting the highest intracellular Zn2+ ion content.

TSQ intracellular Zn2+ probe detection of temporal changes in Zn2+ release from ZnO and H-ZnO in cells. White arrow indicates fluorescence signal of intracellular zinc ion release. To further clarify intracellular Zn2+ release, the TSQ (30 µM; 30 min) probe was used to detect fluorescence in L929 cells. After 15 min of ZnO treatment, intracellular Zn2+ levels significantly increased and continued to rise over time.

Representative images of TSQ (10 μM; 30 min), a fluorescent Zn2+ probe, in Caco-2 cells. The results showed that Zn and I3C combined treatment significantly increased blue fluorescence intensity.

Guide (The following is our recommended protocol. This protocol is for reference only and should be modified according to your specific needs).
    1. Solution preparation[4]
    1.1 Preparation of stock solution
    Solvent: DMSO
Concentration: 1-30 mM (optimized according to the experiment).
Storage: Store at -20°C or -80°C in dark after aliquoting. Avoid repeated freezing and thawing.
    1.2 Preparation of working solution
Dilute to 1-30 μM with DPBS or other buffers.
Note: The working solution should be prepared and used immediately. Keep it away from light.

    2. Cell staining
2.1 Cells are grown on glass coverslips coated with Collagen, rat tail (HY-NP010).
2.2 Rinsed three times with Dulbecco's phosphate buffered saline.
2.3 30 μM TSQ in DPBS, mix well and incubate at 37°C for 30 minutes.
2.4 After the stained cells are rinsed three times with DPBS.
    2.5 Fluorescence microscopy detection (Ex/Em = 334/495 nm).

MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.

    Help & FAQs
    • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
