ATTO 665
ATTO 665 is a near-infrared amine-reactive fluorescent NHS ester dye that can covalently label chitosan for in vivo tracking, or label phage coat proteins for validation of fungal cell binding, and can be used for background monitoring in real-time PCR. ATTO 665 can be used for biomolecular fluorescent labeling and in vivo imaging studies.

Biological Activity
Description
In Vitro
Phage clones labeled with ATTO 665 (1013 phage particles; 15 min) selectively bind to their respective Candida species, producing strong surface-associated fluorescence as observed by fluorescence microscopy[4].
ATTO 665 (15 mg dissolved in 200 µL DMSO; 4 h) is conjugated to chitosan to afford 3.2 g of blue ATTO 665-labeled chitosan in 82% yield[1].
ATTO 665 (0.03 μM; 95°C for 30 s, followed by 40 cycles of 96°C for 1 min and 55°C for 45 s) is added to the second-round real-time PCR premix for background monitoring[2].
ATTO 665 serves as a single energy acceptor in single-molecule FRET experiments using light-harvesting nanoparticles, achieving a FRET efficiency of 51% and enabling single DNA/RNA detection through target-displacement color switching with an LOD of 2 pM[3].
ATTO 665 (2.5 mg/mL; 60 min) is used to label M13 phage coat proteins in PBS-NaHCO3 at pH 8.3 at room temperature, targeting approximately 300 dye molecules per phage[4].
ATTO 665 (5 × 10-7 M; 0-20 ns) has a ground-truth lifetime of 2.9 ns and is resolved by Kernelizing-PARAFAC-ALS, yielding a 15-point sampled lifetime of 2.8 ns with a concentration spectral metric of 0.88, whereas PowerSlicing gives 2.64 ns and 0.83[5].
MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.
